As referenced in numerous threads from last season, the writers of Lost stated that the comic Walt was reading was placed very intentionally and would have some bearing on the story of Lost (the comic was a Spanish translation of "Green Lantern/Flash: Faster Friends # 1").
Now that some character devlopment has taken place and we partially know the contents of the hatch, I'd like to theorize on a possible parallel between Desmond and the antagonist of the comic.
Here (http://www.glcorps.org/alien-x.html) is an excellent summary of Alien X's role in the comic and some background on where I'm about to go.
What if Desmond was a misunderstood doctor/scientist in training that discovered some sort of genetic anomaly, psychic power, magnetic phenomenon, space-time tear, etc that he devoted his life to studying and even built a hidden, underground research facility in which to study it? Stocked with the comforts of any late-70's home, he was content to live eccentrically and alone. His intentions could have been peaceful and benevolent, but once the government learned of his experiments they captured him and ran experiments of their own on him.
At some point, Desmond escaped and "raced around the world" to get back to his underground research facility, but this time, infected with an incurable disease, he is angry and looking for revenge. Enter Wally West and Kyle Rayner...er, Jack and Locke. They realize that Desmond X is out to destroy the world with some sort of doomsday device (sorry, couldn't stretch it enough to include the summoning of alien hordes) and once they overcome their differences, fight together to stop his evil intentions.
The 1st and 2nd generation Flash and Green Lantern's in the comic get a little confusing, but it could be possible that Desmond has The Others under some sort of mind control to serve the same purpose as the 1st generation superheroes.
A stretch, maybe...but nevertheless, a theory.
